Message	tire without taking it off the rim is not worth it/safe. Now, balancing... the wheel will need to be rebalanced but not because of the plug; that has little to no effect on the balance. The tire not being in the same position as it was before with the rim does require it to be balanced. Anyways make sure you use a plug/patch and you should be good. Check out the links below.
